师兄，为什么我的细胞培养液是紫红色？

实验室新来了一个师弟，老板让其从养细胞开始。这个小师弟是个夜猫子，总喜欢深夜做实验。一天早上：
小师弟： 湿胸，昨天传的细胞没贴壁，培养液变成紫红色的了。是不是你给我的那皿细胞是不好的？
科研狗：你操作没毛病？
小师弟：绝对没毛病，严格遵循你的指示。为了怕孵箱细胞太多，我特意单独放在另外一个孵箱里。
科研狗：等等，另外一个孵箱？没有接通CO2啊

今天我们来聊一聊培养液中的颜色，一般情况下，我们购买回来的DMEM培养基都是鲜红色的，如下图：

但是，作为老司机的你肯定也见过如下颜色的：

小师弟： 师兄，你见过赤橙黄露青蓝紫的吗？
科研狗：…

1. 为什么这个DMEM培养基是鲜红色的？
这个问题可以从如下两个方面来回答：
业余回答：
A. 它总要有一个颜色吧，如果是绿色的或者你会问他为什么是绿色的
B. 红色的看上去有食欲，细胞自然喜欢吃了。你想想，给你一堆绿了吧唧的水果和给你一堆鲜红的水果，你喜欢吃哪一种？

专业回答：
培养基的颜色主要是酚红来显示的，酚红指示颜色非常灵敏，p值范围为6-8，颜色由黄变为紫红。酚红不是必须的，在有些情况下甚至是多余的，酚红有类似固醇类激素的作用，如果涉及到固醇类激素相关的细胞培养，最好不要添加。但是为了培养方便，能让我们直观地感觉培养液的状况，加入酚红后，如果培养液偏酸了，就显示偏黄色，偏碱性了就显示偏紫色。

2. 为什么培养皿放在一个没有开启CO2的孵箱里面，培养液颜色偏紫色？
一般的DMEM使用的缓冲体系是NaHCO3， 初中的时候应该见过如下的方

2NaHCO3=Na2CO3+CO2+H2O

在没有CO2的孵箱中，培养液中的CO2一直溢出，碳酸变少了，溶液变碱性了，就偏紫色了。 同时我们打开了很久的DMEM瓶里面的培养液也会慢慢偏紫色，也就是这个道理。

3. 什么时候溶液会变为黄色？
这个简单，一般污染细菌的时候或者长时间不换液，溶液变为黄色主要是酸性物质太多，细菌和细胞过度增长都会产生酸性物质，溶液会变为黄色。

4. 培养基偏紫色了还能用不？
可以将该偏紫色的培养基置于CO2孵箱中，拧松瓶盖。一段时间之后你会发现颜色变为鲜红色了，那么可以继续接着用。但是不要使用过长时间未用完并且变色的培养液，置于空气下氧化之后培养基成分可能已发生了变化。

5. 哪些情况下不得使用酚红？
如前面所说的酚红有类似固醇类激素的作用，如果涉及到固醇类激素相关的细胞培养，不要添加。另外在做流式细胞的时候，酚红会对结果产生影响，因此也不要添加。

6. 有没有不受CO2影响的培养基？
有，如果缓冲体系不是NaHCO3，而是其他体系如HEPES，就不受CO2的影响。

7. 常用的培养基有哪些？
MEM（Minimum Essential Medium）: 也称最低必须培养基，它仅含有12种必须氨基酸、谷氨酰胺和8种维生素。可以广泛应用到各种细胞培养。

DMEM：分为高糖和低糖，碳酸氢钠缓冲体系，所以需要CO2。
高糖的适合于高密度悬浮细胞培养，适用于附着性较差，但又不希望它脱离原来生长点的细胞。 低糖的适合于生长速度较快，附着性较差的肿瘤细胞培养。

DMEM/F12：F12 培养基成分复杂，含有多种微量元素，和DMEM以1：1结合，称为DMEM/F12培养基（DME/F12medium），作为开发无血清配方的基础，以利用F12含有较丰富成分和DMEM含有较高浓度的营养成分为优点。该培养基适用于血清含量较条件下哺乳动物细胞培养。

RPMI-1640：主要用于悬浮细胞培养，广泛用于哺乳物，特殊造血细胞，正常或者恶性增生的白细胞，杂交瘤细胞。 另外K-562， L-60、urkat、IM-9 等淋巴细胞、T细胞淋巴瘤细胞也可以使用。

McCoy’s 5A : 主要用于肉瘤细胞的培养， 还可以用于骨髓、皮肤、肺等原代培养，用于作组织活检培养、一些淋巴细胞培养以及一些难培养细胞的生长支持。比如HT29，BHL100等上皮细胞。

IMDM（Iscove’s Modified Dulbecco Medium）：主要用于红细胞和巨噬细胞前体培养。培养液中含有微量元素硒以及额外的氨基酸和维生素、丙酮酸钠以及HEPES。

M199 Medium 是具有确定化学成分的细胞培养液，主要用于鸡胚成纤维细胞培养，培养时必须辅助血清才能长期培养。

L-15(Leibovitz Medium) 适用于在CO2缺乏的情况下培养肿瘤细胞株，用于快速增值瘤细胞的培养。

Ham’s F-10: 适用于小鼠细胞以及人的二倍体细胞的培养

Ham’s F-12: 适用于单细胞培养或者克隆培养， 是无血清培养基中常用的基础培养基。

MCDB131： 适用于培养内皮细胞。

Opti-MEM I Reduced Serum Media: 适用于培养造血细胞。
