德国亚琛大学报告了一种智能盐桥设计策略，同时提高模型酶枯草杆菌脂肪酶A（Bacillus subtilis lipase A，BSLA）（WT：PDB ID:1i6w）的有机溶剂耐受性和热稳定性。结合对3450个酶突变体的盐桥形成与断裂的分析后，发现BSLA表面形成的不利盐桥可能是导致酶的有机溶剂耐受性下降的原因，综合实验研究和36个体系的分子动力学模拟，获得了四个有益突变体，对三种有机溶剂的耐受性提高了7.6倍（D64K/D144K），同时热稳定性提高了137倍，半衰期提高了3.3倍。分子动力学模拟显示，蛋白局部柔韧性和水化层的增强共同决定了酶在有机溶剂中的耐受性和50-100℃下热稳定性的增加。盐桥的重新设计为蛋白质工程师提供了一个可能是通用的方法来设计OS和/或抗热脂肪酶和其他α/β水解酶。

作者将仅在有机溶剂中形成的盐桥定义为“不利盐桥”，认为这样的盐桥限制了酶的灵活性与活性。通过去除表面的不利盐桥，酶的有机溶剂耐受性和热稳定性可以得到显著改善。

使用MD对BSLA WT模拟时，观察到在有机溶剂中，形成了三对水相中不存在的盐桥，分别是D34-K35、K61-D64和K112-D144中三个“不利盐桥”的位置。盐桥在极性脂肪酶主要起稳定作用，存在于酶的表面。可以观察到三个盐桥的突变体组合对于有机溶剂的耐受性都有提高。

D64K/D144K和D34K/D64K/D144K的组合突变在高浓度有机溶剂下仍表现出显著的活性；几乎所有的突变体在<15% (v/v) DOX、<20% (v/v)DMSO和<6% (v/v)TFE的有机溶中有被激活的现象。

本文还研究了5个纯化突变体的耐热性。野生型在50-100℃热处理1小时后失去了>90%的活性。相比之下，除 D34K/D64K/K112E/D144K外，所有突变体的热阻在40-100℃的整个温度范围内均显著增加。值得注意的是，D34K/D144K 在100℃ 时的残留活性比野生型高出137倍。有趣的是，在高温(> 70℃)条件下，BSLA 发生了二次和/或三次取代反应，残留活性升高到70% 左右。这一现象表明，在<70℃和>75℃时，变性方式可能不同。在达到变性温度(≈70℃)时，蛋白质可能展开成易于不可逆聚集和沉淀的中间体。
